A lo largo de su carrera, Charles Lane participó en muchas películas y programas de televisión. Es conocido por aparecer en varias películas del famoso director Frank Capra, como Caballero sin espada, Arsenic and Old Lace y ¡Qué bello es vivir!.

Lane comenzó trabajando como vendedor de seguros. Luego, decidió seguir su pasión por la actuación en el teatro de Pasadena.

En 1929, el actor y director Irving Pichel le sugirió que empezara a actuar. Cuatro años después, Lane fue uno de los fundadores del Screen Actors Guild, un grupo importante para los actores.
